Definition

Imagine building a long chain out of many small, identical building blocks. In the world of biology, proteins are these long chains, and their building blocks are called amino acids. To connect these amino acid building blocks together, a special type of chemical 'glue' is used, and that glue is what we call a peptide bond.

Let's break it down: Every amino acid has two key functional groups: an amino group (NH2-\text{NH}_2) and a carboxyl group (COOH-\text{COOH}). When two amino acids want to join hands, the carboxyl group of one amino acid comes close to the amino group of another.

In this interaction, the hydroxyl group (OH-\text{OH}) from the carboxyl group and one hydrogen atom (H-\text{H}) from the amino group combine to form a molecule of water (H2O-\text{H}_2\text{O}). This water molecule is then released, and a new bond forms between the carbon atom of the carboxyl group and the nitrogen atom of the amino group.

This newly formed CONH-\text{CO}-\text{NH}- bond is the peptide bond.

This process is called a 'condensation reaction' or 'dehydration synthesis' because a water molecule is removed. Once formed, this bond is very strong and stable. It's not just a simple single bond; it actually has characteristics of both a single and a double bond due to something called 'resonance'.

This partial double bond character makes the peptide bond quite rigid and planar, meaning the atoms directly involved in the bond and those immediately attached to them tend to lie in the same flat plane.

This rigidity is incredibly important because it restricts the rotation around the peptide bond, which in turn dictates how the protein chain can fold into its specific three-dimensional shape, ultimately determining its biological function.

Without peptide bonds, there would be no proteins, and without proteins, life as we know it would not exist.
